Abstract: Topical herbal formulations have emerged as promising alternatives to synthetic pain-relieving preparations owing to their therapeutic efficacy, ease of application, and reduced systemic adverse effects, wherein naturally derived bioactive compounds such as thymol, menthol, and camphor are recognized for their analgesic, anti-inflammatory, antimicrobial, and soothing properties. The present invention provides a novel herbal medicated balm comprising thymol, menthol, camphor, medicated herbal oil, and beeswax formulated as a semi-solid composition for external application. The developed formulation was evaluated for its anti-inflammatory, antimicrobial, cytocompatibility, and physicochemical properties and demonstrated concentration-dependent anti-inflammatory activity with inhibition values ranging from 25.04% to 83.50%. The formulation further exhibited antibacterial activity against Escherichia coli and Bacillus subtilis and antifungal activity against Candida albicans and Candida tropicalis. Cytotoxicity assessment using Human Vero cells revealed a viability of 98.67%, indicating excellent biocompatibility and negligible toxicity. Additionally, the balm exhibited desirable physicochemical characteristics including a skin-compatible pH of 5.8, smooth texture, satisfactory absorbency, ease of washability, and absence of skin irritation. The developed herbal balm therefore represents a safe, stable, and effective topical therapeutic composition for the management of pain, inflammation, muscular stiffness, headache, and joint discomfort.
