Abstract: ABSTRACT NOVEL PYRAZOLO[3,4-D] PYRIMIDINE DERIVATIVES AND PROCESSES FOR PREPARATION THEREOF The present invention relates to novel pyrazolo[3,4-d]pyrimidine derivatives of Formula (I), and pharmaceutically acceptable salts, solvates, hydrates, stereoisomers and polymorphic forms thereof, useful as Heat Shock Protein 90 (HSP90) inhibitors. The compounds comprise substituted pyrazolo[3,4- d]pyrimidine scaffolds bearing substituted benzamide moieties, wherein the substituents enhance binding affinity toward the N-terminal ATP-binding domain of HSP90 and provide potential anticancer activity. The invention further provides processes for preparing the compounds of Formula (I), comprising reacting an arylhydrazine with ethoxymethylenemalononitrile to obtain an aminopyrazole intermediate, cyclizing the intermediate with formamide to obtain a pyrazolo[3,4- d]pyrimidine intermediate, and coupling the intermediate with a substituted benzoyl chloride or activated substituted benzoic acid in the presence of a coupling reagent and an organic base. The compounds exhibit favorable physicochemical and biological properties and are useful for treating or preventing HSP90-mediated disorders, including cancer.
