Abstract: The present invention The present invention relates to a topical vesicular gel formulation of tofacitinib for localized rheumatoid arthritis therapy. The formulation comprises tofacitinib encapsulated in lipid-based nanovesicles selected from liposomes, ethosomes, or a combination thereof, uniformly dispersed in a Carbopol-based gel matrix. The liposomal vesicles comprise phospholipid and cholesterol, while the ethosomal vesicles comprise phospholipid, cholesterol, ethanol, propylene glycol, and aqueous phase. The nanovesicles have nanoscale particle size, improved drug entrapment, and spherical morphology. The gel matrix comprises Carbopol 940, triethanolamine, glycerin, water, and optionally propylene glycol, providing suitable viscosity, spreadability, pH, and physical stability. The formulation enhances skin permeation and sustained localized delivery of tofacitinib compared with raw drug. Ethosomal gel provides superior permeation due to ethanol-mediated stratum corneum fluidization and vesicle deformability. The invention provides a stable, patient-friendly topical formulation useful for reducing localized inflammation in rheumatoid arthritis while minimizing systemic exposure.
