Abstract: ABSTRACT “NYSTATIN-LOADED SELF-MICROEMULSIFYING DRUG DELIVERY SYSTEM FOR ENHANCED BIOAVAILABILITY” The present invention relates to a pharmaceutical composition comprising a -loNyadstatied nself- microemulsifying drug delivery system (SMEDDS) for enhancing the solubility, dissolution, and bioavailability of Nystatin. The composition includes an oil phase, at least one surfactant, and at least one co-surfactant forming an isotropic pre-concentrate that spontaneously generates a fine oil-in-water microemulsion upon contact with gastrointestinal fluids. The resulting microemulsion, having droplet sizes typically below 100 nanometers, maintains Nystatin in a solubilized state irrespective of pH variations and facilitates improved absorption through mechanisms including diffusion, endocytosis, and lymphatic transport. The formulation reduces variability in pharmacokinetics and enhances therapeutic efficacy compared to conventional dosage forms. The invention provides a stable, scalable, and patient-compliant oral delivery system suitable for improving the performance of poorly water-soluble antifungal drugs.
