Abstract: The present invention relates to p53-SenoDef-v2, a computationally engineered recombinant human TP53 protein variant comprising six targeted point mutations S15D, S20D, S46D, K120Q, E346A, and K382R that simultaneously preserve apoptotic transcriptional activity, retain ferroptosis-inducing capacity via SLC7A11 repression, and ablate senescence-promoting transcriptional activity via p21 elimination and mTORC1-SASP disruption. The invention further relates to a lipid nanoparticle delivery system beta-SitoLPN-v2 comprising DPPC at 45-55 mol percent, beta-sitosterol at 25-30 mol percent for mTORC1-SASP suppression via lipid raft disruption, DOPE at 15-20 mol percent for pH-responsive endosomal escape, DSPE-PEG2000, p28 cell-penetrating peptide conjugated to DSPE at 1-2 mol percent for cancer-selective delivery and post-delivery p53 stabilisation via HDM2-independent proteasomal inhibition, and folate-DSPE at 0.5-1 mol percent for folate receptor alpha targeted delivery. The invention further relates to a codon-optimised mRNA encoding p53-SenoDef-v2 optionally with N1-methylpseudouridine modification encapsulated in beta-SitoLPN-v2 as the scalable commercially preferred mRNA-LNP therapeutic embodiment. The combined product p53-SenoDef-LPN- v2 is the first-in-class triple-mechanism non-viral senescence-decoupled p53 therapeutic covering protein delivery, mRNA delivery, phytosterol SASP suppression, cancer-selective CPP delivery, and folate receptor targeting in a single integrated platform. The invention encompasses the engineered protein sequence, all six mutations and their combinations, the mRNA encoding the protein, the lipid nanoparticle formulation, and methods of use for cancer therapy, senescence prevention, progeroid syndromes, osteoarthritis, and Long COVID sequelae.
