Abstract: ABSTRACT [0033] The present study focused on the development and evaluation of curcumin-loaded hydrogel formulations to enhance wound healing activity. Wound healing is a complex, multi-stage physiological process involving inflammation, proliferation, and tissue remodeling, necessitating effective and patient-friendly topical delivery systems. Curcumin, a natural polyphenolic compound derived from turmeric, was selected due to its well-established anti-inflammatory, antioxidant, and antimicrobial properties. Aloe vera was incorporated to improve soothing effects, hydration, and epithelial regeneration.Four hydrogel formulations (F1–F4) were prepared using Carbopol 940 as the gelling agent, along with propylene glycol, ethanol, glycerine, and suitable preservatives. The formulations differed in concentrations of curcumin and aloe vera to evaluate their influence on formulation performance. [0034] All prepared hydrogels were subjected to physicochemical evaluation, including pH, viscosity, swelling index, gelation time, and spreadability. The pH values (5.5–7.4) were within the acceptable physiological range, ensuring skin compatibility. The formulations exhibited suitable viscosity for effective retention at the application site, good swelling capacity, rapid gelation, and satisfactory spreadability. Among them, F3 showed optimal physicochemical characteristics. In vivo wound healing studies using Wistar rats demonstrated enhanced wound contraction in all treated groups compared to the control. The standard (povidone iodine) showed faster healing, while F3 exhibited the highest wound closure rate, likely due to increased aloe vera content. F4 also showed consistent healing with balanced properties.
