MUMBAI, India, Aug. 17 -- Intellectual Property India has published a patent application (202621084924 A) filed by Parul Institute Of Ayurved, Parul University on July 10, 2026, for Polyherbal Injectable Formulation With Detoxified Aconitum Ferox For Local Anesthesia.

Abstract: A polyherbal injectable formulation for local anesthesia comprises standardized hydro-alcoholic extracts of detoxified Aconitum ferox, Anacyclus pyrethrum, and Myristica fragrans in a 2:1:1 weight ratio, formulated in a sterile isotonic aqueous vehicle. Aconitum ferox undergoes Ayurvedic Shodhana detoxification using cow's urine and cow's milk at 90–95°C for approximately 3 hours, converting toxic aconitine alkaloids into safer derivatives while preserving anesthetic activity. Each herb is extracted separately via hydro-alcoholic Soxhlet extraction (ethanol:water 50:50 v/v) for 7 cycles. The combined extracts are processed by homogenization (5000–6000 rpm, 5–15 minutes) and ultrasonication (2 minutes), producing a stable nano-dispersion with 100–150 nm particle size, polydispersity index below 0.3, and zeta potential approximately –22 mV. The formulation is sterile-filtered (0.22 µm), pH-adjusted (6.8–7.2), and aseptically filled into Type-I borosilicate glass ampoules. In vivo evaluation demonstrates dose-dependent reversible peripheral nerve conduction blockade with efficacy comparable to 2% lignocaine, approximately 60-minute anesthetic duration, NOAEL exceeding 10 mg/mL, and pharmaceutical stability for at least six months under ICH storage conditions.
