Abstract: Process for the biosynthesis of volatile disulfide compounds from in vitro induced Ferula assafoetida callus culture Abstract The characteristic aroma of Ferula assafoetida (Heeng) is mainly attributed to sulfur-containing compounds released by the dried latex (oleo-gum resin) exuded from its taproots. Hence, these organosulfur derivatives i.e. disulfide compounds are considered as chemotaxonomic markers for Ferula species. In this invention, callus cultures were induced from F. assafoetida leaves on standardized Murashige and Skoog medium supplemented with 2,4-dichlorophenoxyacetic acid and Kinetin. In order to assess the scalability, in vitro process parameters like medium strength and sucrose concentration including process parameters (agitation & inoculum density) were optimized in liquid cultures. Further, the biosynthesis of disulfide derivatives was achieved with the treatment of calli with different precursors in appropriate vessel and drying condition. The Gas Chromatography-Mass Spectrometry - Headspace analysis showed a significant amount of dimethyl disulfide (60.0%) and dimethyl trisulfide (18.0%) in leaf-induced F. assafoetida calli. Thus, the present invention demonstrated a regenerative process as a substitute to field cultivation for producing volatile disulfide compounds in short cultivation period.
