MUMBAI, India, July 31 -- Intellectual Property India has published a patent application (202411102236 A) filed by National Institute Of Pharmaceutical Education And Research Niper on December 23, 2024, for Process For The Preparation Of Metaxalone.

Abstract: The present invention relates to a process for the synthesis of metaxalone having formula (I) in an efficient, safe and simple manner, which is free from the problems encountered in the methods described in the prior art. The process of the present invention is a two-step process for the preparation of metaxalone. Accordingly, the process of present invention involves reaction of m-xylenol with epichlorohydrin, followed by ring opening of the epoxide intermediate with aqueous ammonia to give 1-amino-3-(3,5-dimethylphenoxy)propan-2-ol, which upon catalytic cyclization with diphenylcarbonate (DPC) in the presence of catalytic amount of TBAF gives metaxalone compound of formula (I).
