Abstract: ABSTRACT PRODRUGS OF ISONIAZID WITH ENHANCED PLASMA EXPOSURE AND METHOD OF DEVELOPMENT THEREOF The present disclosure relates to prodrugs of isoniazid. The disclosed prodrugs comprise carbamate, urea, and amide derivatives of isoniazid, wherein the terminal –NH2 group of isoniazid is chemically modified to form prodrug structures capable of releasing isoniazid under biological conditions. The synthesized prodrugs designated as PD-1 to PD-42 exhibit chemical stability under selected conditions and demonstrate biological lability in plasma, resulting in the release of isoniazid. The disclosure also includes pharmacokinetic and biological evaluations illustrating in vivo conversion of selected prodrugs to isoniazid. The disclosed compounds provide multiple methods and approaches for synthesizing isoniazid prodrugs suitable for further pharmaceutical development. FIG. 2
