Abstract: The present invention relates to a QbD-guided development of taxifolin-loaded ethosomes (TAX-ETH) for enhanced dermal delivery and improved therapeutic management of psoriasis. The ethosomal formulation was optimized using a Box–Behnken design and prepared using soya lecithin and ethanol by a cold method to obtain vesicles with optimized particle size, polydispersity index, entrapment effxiciency, and stability. The optimized formulation exhibited nanosized vesicles, high drug entrapment, controlled drug release, and improved skin permeation and retention compared to conventional formulations. Characterization studies including TEM, SEM, FTIR, DSC, and XRD confirmed successful incorporation of taxifolin within the ethosomal vesicles. The TAX-ETH gel demonstrated suitable topical properties and significantly reduced psoriasis severity in in vivo studies with effects comparable to clobetasol. Network pharmacology analysis further identified multiple therapeutic targets and signaling pathways associated with the anti-psoriatic activity of taxifolin, thereby establishing the formulation as a promising nanotechnology- based dermal delivery system.
