Abstract: The present invention relates to a rapid assay to detect antigenic lipids in a test tube from tuberculosis patients’ serum and a method thereof. Tuberculosis, caused by Mycobacterium tuberculosis, is characterized by a lipid-rich cell wall containing mycolic acids that exhibit strong antigenic properties. The invention discloses a novel immunoassay based on antigen antibody agglutination for the detection of mycobacterial lipid antigens, particularly mycolic acids, using antibodies present in patient serum. The method overcomes lipid solubility challenges while preserving antibody integrity in the presence of organic solvents. Detection is achieved through visible agglutination without the need for enzymatic labels or specialized instrumentation. The assay is simple, rapid, cost-effective, and adaptable for clinical diagnostics and research applications, offering a practical platform for tuberculosis screening and future lipid-based diagnostic development.
