Abstract: The present invention provides a rapid, simple, accurate, and cost-effective Reverse Phase High Performance Liquid Chromatography (RP-HPLC) method for the simultaneous estimation of Domperidone and Cinnarizine in pharmaceutical bulk and dosage forms. Chromatographic separation is achieved on a C18 Sunfire column (5 µm, 250 mm × 4.6 mm) using a mobile phase of orthophosphoric acid buffer and acetonitrile (60:40 v/v) in isocratic elution mode at a flow rate of 1.0 ml/min with PDA detection at 226 nm. The retention times of Domperidone and Cinnarizine are 2.52 minutes and 5.18 minutes respectively. Linearity is established in concentration ranges of 1 to 5 µg/ml for Domperidone (R² = 0.9999) and 5 to 25 µg/ml for Cinnarizine (R² = 0.9996). The limits of detection are 0.3325 µg/ml and 0.615 µg/ml for Domperidone and Cinnarizine respectively. The method is validated for accuracy, precision, robustness, and ruggedness in accordance with ICH guidelines and is suitable for routine quality control analysis
