Abstract: The present disclosure provides devices and methods for separation of capture particles into wells of an array of wells and sealing the wells with a hydrophobic liquid. In certain embodiments, the methods of the present disclosure involve detecting presence of a signal from the array of wells, wherein the presence of the signal is indicative of the presence of an analyte immobilized on capture particles. In certain embodiments, the device for separation of capture particles into wells of an array of wells and sealing the wells with a hydrophobic liquid includes protrusions at an opening configured to prevent the hydrophobic liquid from reaching the opening before the hydrophilic liquid is removed.
