Abstract: Embodiments of the present disclosure relate to a semi-quantitative diagnostic kit (100) and method (200) for assessing Vitamin A deficiency in a biological sample. The kit (100) includes a lateral flow assay strip (102), a nitrocellulose membrane, an absorbent pad, a sample treatment buffer and a reference classification unit. The sample receiving pad (104) is configured to receive the biological sample. The conjugate pad (110) containing a detectable conjugate includes one or more retinol- binding protein (RBP)-specific antibodies conjugated to signal-generating nanoparticles. The absorbent pad is configured to maintain capillary flow through the lateral flow assay strip (102). The sample treatment buffer is configured to facilitate migration of the biological sample through the lateral flow assay strip (102). reference classification unit including a visual intensity chart correlating signal intensity at the test line (106) with a plurality of Vitamin A status categories derived from retinol- binding protein concentration ranges.
