Abstract: Title: Smart Chitosan Gated Ca-PSZ Disc as Drug Delivery System and preparation Thereof Peri-implantitis, biofilm-mediated infection that generates a localized acidic microenvironment (pH 5.5) is a primary cause for dental implant failure, the current drug delivery systems are often limited by nonspecific drug release that remains less response to infection. This invention successfully developed and validated a novel, biocompatible calcium oxide-partially stabilized zirconia (Ca-PSZ) composite featuring a pH- responsive "smart gate" fabricated from chitosan cross linked with citric acid. The porous scaffold was produced using cost-effective NaCl particulate-leaching and twostage sintering method, achieving an interconnected architecture confirmed by 34.6% mass reduction post-leaching and visual radial diffusion studies. Quantitative diffusion analysis demonstrated the systems autonomous gating efficacy by exhibiting significant decrease in drug release at neutral physiological conditions (pH 7.4), but exposure to simulated infected environment (pH 4.0-5.0) triggered protonation of chitosan backbone, resulting in a controlled release of 43.30 % of chlorhexidine pay load within 48 hours. The biological validation through MTT assays confirmed 88.60% cell viability even at maximum concentration, while antimicrobial testing yield 17 mm zone of inhibition against streptococcus mutans, comparable to clinical standards. These results establish a functional proof-of-concept for a self-regulating, metal-ftee dental implant that provides high demand antimicrobial protection while ensuring the long-term health of surrounding alveolar tissues.
