Abstract: The present invention discloses a self-oxygenating galactose-functionalized manganese-porphyrinic metal-organic framework nanoplatform designated AteBev@Gal- PM for synergistic immuno-photodynamic therapy of hepatocellular carcinoma. The nanoplatform comprises a PCN-224(Mn) core exhibiting catalase-like activity for in situ oxygen generation from endogenous hydrogen peroxide, a PEG coating bearing galactose targeting ligands for ASGPR-mediated hepatocellular carcinoma targeting, and surface-conjugated atezolizumab and bevacizumab monoclonal antibodies for concurrent immune checkpoint blockade and vascular normalisation. Upon 660 nm irradiation, the nanoplatform generates singlet oxygen under hypoxic conditions while photodynamic therapy induced immunogenic cell death synergistically amplifies the anti-tumour immune response. The invention achieves a tumour inhibition rate exceeding 80 percent in animal models with favourable biosafety. The invention further provides scalable synthesis, tuneable ligand density, defined irradiation protocols, alternative quinone co-loading, and subtype-specific dosing schedules.
