Abstract: A two-stage enzyme-substrate based paper strip sensor has been developed for the rapid detection of Aflatoxin B1 (AFB1), leveraging Bacillus stearothermophilus- based spores and a-glucosidase as the marker enzyme. Screening of six enzymes identified a-glucosidase as the most sensitive, producing a distinct color change on functionalized paper strips within 30 minutes. Spore-based enzyme activity was optimized by adjusting spore concentration, substrate volume, incubation temperature, and reaction time, with 65°C and 30 minutes identified as optimal conditions. The assay demonstrated a limit of detection (LOD) of 10 ppb for AFB1, meeting regulatory thresholds for feed and food safety. Paper strips, fabricated using Whatman Grade 3 filter paper, exhibited reliable enzyme-substrate reactions, validated through extraction efficiency, inhibition assays, and chromogenic detection. Validation using ELISA and HPLC-FD confirmed the assay's accuracy, with results aligning closely with established analytical methods. Field trials on 120 feed and food samples demonstrated the assay's specificity, detecting AFB1 without cross- reactivity to other contaminants such as pesticides, heavy metals, and antibiotics. This novel paper strip biosensor provides a rapid, cost-effective, and field-deployable method for AFBI detection in diverse matrices, ensuring compliarice with safety standards and supporting efforts to mitigate aflatoxin-related risks in animal feed and food products.
