Abstract: Rheumatoid arthritis is a chronic autoimmune inflammatory disorder characterized by persistent inflammation, synovial hyperplasia, and progressive destruction of cartilage and bone. Although conventional anti-arthritic drugs provide symptomatic relief, their long- term use is associated with several adverse effects, necessitating the search for safer and effective plant-based alternatives. Helianthus annuus L. (sunflower) seeds are rich in bioactive lipophilic constituents such as unsaturated fatty acids, tocopherols, phytosterols, and flavonoids, which possess anti-inflammatory and antioxidant properties. The Objective of present study aims at evaluate the in vitro anti-arthritic activity of the petroleum ether extract of Helianthus annuus L. seeds using protein denaturation inhibition and human red blood cell (HRBC) membrane stabilization assays. Petroleum ether extract of Helianthus annuus L. seeds was prepared by Soxhlet extraction. The anti-arthritic activity was assessed by inhibition of protein denaturation and HRBC membrane stabilization methods at different concentrations of the extract. Diclofenac sodium was used as the standard reference drug. The percentage inhibition of protein denaturation and membrane stabilization was calculated and compared with the standard. The maximum percentage inhibition of protein denaturation and percentage protection of membrane stabilisation for Helianthus annuus L extracts were found to be 75.14 ± 0.154 % and 84.13 ±0.018% respectively at a dose of 200µg/ml.
