Abstract: Advanced wound-care systems capable of providing controlled therapeutic delivery and enhanced tissue regeneration are increasingly required for the effective management of acute and chronic wounds. The present invention relates to a stimuli-responsive hydrogel patch loaded with Calotropis gigantea extract for accelerated wound repair. The hydrogel patch comprises a biocompatible polymeric network capable of responding to wound-associated physiological conditions and facilitating the sustained release of therapeutically active phytoconstituents. Bioactive compounds present in Calotropis gigantea contribute antimicrobial, antioxidant, and anti- inflammatory activities that assist in controlling infection, reducing oxidative stress, and promoting tissue regeneration. The hydrogel matrix further provides moisture retention, exudate absorption, and a protective environment favorable for cellular proliferation and wound closure. The developed patch demonstrates excellent biocompatibility and enhanced wound-healing performance, making it suitable for the treatment of acute wounds, chronic wounds, diabetic ulcers, burn injuries, pressure sores, and other skin lesions. The invention offers a safe, effective, and plant-based wound management platform with improved therapeutic efficacy and patient compliance.
