Abstract: The present invention relates to a stimuli-responsive nanoparticle composition for targeted and controlled drug delivery in biological systems. The disclosed invention comprises a multifunctional nanoparticle core configured to encapsulate or conjugate one or more therapeutic payloads including anticancer agents, antimicrobial compounds, nucleic acids, proteins, peptides, vaccines, and regenerative therapeutic molecules. The nanoparticle surface is functionalized with targeting ligands such as antibodies, aptamers, peptides, receptor-binding molecules, or biomimetic structures for selective localization at disease-specific tissues or cells. The composition further incorporates a stimuli-responsive polymeric coating capable of regulating controlled therapeutic release in response to physiological or externally induced triggers including pH variation, temperature fluctuation, enzymatic activity, oxidative stress, magnetic stimulation, ultrasound exposure, or light irradiation. Stealth stabilization layers are additionally integrated to reduce immune recognition and prolong systemic circulation. The invention may further include diagnostic imaging agents and biosensing components for theranostic applications and real-time therapeutic monitoring. The disclosed nanoparticle platform improves therapeutic precision, drug bioavailability, circulation stability, and treatment efficacy while minimizing systemic toxicity and off-target side effects. The invention further provides scalable manufacturing methodologies and broad applicability in oncology, infectious diseases, cardiovascular therapy, neurological disorders, regenerative medicine, gene delivery, vaccine administration, and precision healthcare systems.
