Abstract: The present invention discloses the synthesis, characterisation, and antimicrobial evaluation of a novel borneol-based chalcone derivative, 3-(4-isopropyl-3- nitrophenyl)-1-(4-(3-(1,7,7-trimethylbicyclo[2.2.1]heptan-2-yl)oxy)propyl)amino)phenyl)prop-2-en-1-one (compound 7B). The compound is synthesised through a three-step route involving O-alkylation of borneol with 1-bromo-3-chloropropane, nucleophilic amination with 4-aminoacetophenone, and Claisen-Schmidt condensation with 4-isopropyl-3-nitrobenzaldehyde. The compound is fully characterised by FTIR and 1H NMR spectroscopy. Molecular docking studies reveal that compound 7B exhibits strong binding affinity against Candida albicans lanosterol 14-alpha-demethylase with GLIDE scores of -10.154 to -10.655, surpassing the standard fluconazole (-6.58), and against Escherichia coli DNA gyrase topoisomerase B with scores of -5.083 to -5.613, comparable to amoxicillin (-5.29). In silico ADMET profiling confirms favourable drug-likeness properties. The compound provides a first-in-class dual-target borneol-chalcone antimicrobial lead.
