Abstract: Abstract: The present invention relates to the synthesis and evaluation of a series of novel 4-arylidene curcumin derivatives prepared using substituted aromatic aldehydes through a Knoevenagel condensation reaction. The synthesized compounds were structurally characterized using standard spectroscopic techniques. Pharmacokinetic and physicochemical properties were evaluated using in silico ADME tools, revealing compliance with Lipinski’s rule of five and favorable drug- likeness. The compounds exhibited high gastrointestinal absorption and low blood-brain barrier permeability. Molecular docking studies demonstrated strong binding affinity with Cyclin-Dependent Kinase 2 (CDK2), indicating potential anticancer activity. Density Functional Theory (DFT) calculations confirmed stability and reactivity. In vitro studies on A549 lung cancer cell lines revealed dose dependent cytotoxicity and G0/G1 phase cell cycle arrest.
