Abstract: ABSTRACT [0012] A synthetic library of orthosteric antagonists targeting the Transient Receptor Potential Ankyrin 1 (TRPA1) channel is disclosed, in which residence times at the orthosteric binding site are systematically tunable to achieve precise temporal control of channel desensitization. Modular structural variations introduced into a core orthosteric scaffold modulate ligand dissociation kinetics while preserving high binding affinity, thereby generating compounds that produce short-, intermediate-, or prolonged-duration functional silencing of TRPA1 after washout. The library is prepared by sequential condensation, cross-coupling, and functional-group attachment reactions that yield discrete, high-purity molecules suitable for cellular and biophysical evaluation. Functional assays confirm that residence time correlates directly with the duration of recovery from desensitization, enabling selection of kinetic profiles matched to specific experimental or therapeutic requirements. The resulting antagonists differ from prior agonist-driven desensitization strategies and from conventional non-tunable TRPA1 blockers by providing deliberate kinetic control. Applications include research tools for dissecting the temporal contribution of TRPA1 activity to sensory signaling and potential therapeutic candidates for conditions involving pathological TRPA1 activation where reversible, time-controlled channel blockade is advantageous.
