Abstract: The present disclosure relates to a system and method to evaluate the efficacy of antimicrobial agents against biofilm-forming microorganisms. The system includes a sample loading unit (101), a standardization unit (102), a media handling unit (103), an inoculation unit (104), a biofilm formation chamber (105) with an environmental control (106), an antimicrobial dispensing unit (107), an optical unit (108), and a processor (202) with memory (204) storing executable instructions. The processor (202) automates sequential biofilm cultivation on a solid culture medium, controlled antimicrobial exposure, image acquisition, and computational analysis of growth parameters including biofilm surface coverage, colony density, inhibition zone diameter, growth kinetics, and relative growth reduction. By comparing treated and untreated regions, the system generates an antimicrobial efficacy profile and susceptibility classification. The disclosure provides clinically relevant, accurate, and reproducible antimicrobial susceptibility testing under biofilm-specific conditions.
