Abstract: A system and method to identify and characterize the antibody isotypes of Mugil cephalus in estuarine fish are disclosed. The system comprises a specimen receiving station, a sample preparation assembly, an antibody extraction unit, an immunoglobulin fractionation module, an isotype detection assembly, a molecular characterization module, a signal acquisition subsystem, and a computational analysis engine. Biological samples obtained from Mugil cephalus are conditioned through microfluidic processing, centrifugation, and filtration operations prior to immunoglobulin isolation within the antibody extraction unit. Purified immunoglobulin constituents are subjected to chromatographic separation, electrophoretic migration, capillary transport, and molecular sieving operations within the immunoglobulin fractionation module to generate separated immunoglobulin fractions. The separated fractions interact with antigen immobilization substrates within the isotype detection assembly to generate fluorescence, chemiluminescent, absorbance, and electrochemical interaction signals. The molecular characterization module generates spectral and sequence information corresponding to the separated immunoglobulin fractions through peptide digestion, ionization, and nucleic acid extraction.
