Abstract: The present invention relates to a herbal hepatoprotective composition comprising an ethyl acetate extract of leaves of Ocimum sanctum Linn. (Tulsi) and a process for the preparation thereof, optionally in synergistic combination with silymarin, for the prevention and treatment of paracetamol-induced hepatic damage in mammals. The said extract is prepared by defatting shade dried leaves with petroleum ether followed by Soxhlet extraction with ethyl acetate, concentration under reduced pressure and drying to constant weight, yielding a free flowing brownish green amorphous powder standardised to a defined phenolic and flavonoid content. The said composition, when administered orally to paracetamol-intoxicated albino rats at two hundred milligrams per kilogram of extract alone or at a half dose combination of one hundred milligrams per kilogram of extract with fifty milligrams per kilogram of silymarin, restores serum proteins, albumin globulin ratio, alkaline phosphatase and serum transaminases towards normal values and produces histological evidence of hepatic regeneration, demonstrating significant hepatoprotection and synergism with silymarin.
