Abstract: The invention discloses an in-silico modified DNA molecule with exceptional thermostability, designed for applications in nanotechnology and molecular engineering. The modification involves the introduction o–fC 5a ’ Cco2’valent linkage within every nucleotide sugar ring of the DNA backbone. All-atom Molecular Dynamics (MD) simulations demonstrate that this intramolecular bridge reinforces the sugar-phosphate backbone, enabling DNA to maintain its double-helical integrity and resist denaturation under extreme thermal stress. The modified DNA preserves standard base-pairing capabilities while offering increased rigidity and reduced structural fraying. This invention provides a robust, thermo-resilient genetic scaffold suitable for DNA origami, high-temperature nanomachinery, and the development of advanced nanoscale biosensors.
