Abstract: Transdermal Drug Delivery formulation of Vortioxetine and its Process for the Preparation Thereof The present invention relates to a transdermal drug delivery system (TDS) comprising Vortioxetine or its pharmaceutically acceptable salts, designed for the treatment of major depressive disorder (MDD) and other central nervous system (CNS) disorders. The Vortioxetine Transdermal Patch (TDS) was developed with aim to reduce hepatic first-pass metabolism, improve patient compliance through reduced dosing frequency, and minimize overdose risk by providing controlled, sustained release of Vortioxetine. The TDS includes a backing layer, a drug- loaded adhesive matrix layer containing Vortioxetine, Duro-Tak 87-2287 pressure-sensitive adhesives, Ethyl oleate as a permeation enhancer, Colloidal Silicon Dioxide as Matrix stiffener, Polyvinyl pyrrolidone as crystal inhibitor and Ethyl acetate as solubilizer. Based on the flux studies, the onset of drug permeation achieved in 3 hours only and maintained the dose till 24 hours. The formulation with 15 sq. cm. patch area is optimized for 10 mg/day strength. And based on the flux, we can predict the delivery of about 20 mg/day for 30 sq. cm. patch and 5 mg/day for 7.5 sq. cm. patch. The system is developed ensuring consistent performance, stability, and improved patient compliance. The invention also provides a method of manufacturing the system.
