Abstract: UNILAMELLAR LIPOSOMAL FORMULATION OF NORCLOMIPRAMINE FOR TREATMENT OF VISCERAL LEISHMANIASIS The present invention provides a unilamellar liposomal formulation of norclomipramine, an active metabolite of clomipramine, for the treatment of visceral leishmaniasis. The formulation comprises norclomipramine encapsulated within unilamellar liposomes, preferably composed of soya phosphatidylcholine, hydrogenated phosphatidylcholine, and cholesterol in a 35:35:30 ratio. Norclomipramine inhibits an essential enzyme, DNA TopoisomeraseIA of Leishmania. Leishmania Topoisomerase IA prevent transcription induced nuclear R-loop accumulation and relaxes negatively supercoiled DNA. Norclomipramine that does not inhibit E. coli TOPIA could inhibit the function of LdTOPIA. Study shows that norclomipramine binds to active site of Leishmania TopoisomeraseIA. Norclomipramine is more potent anti-leishmanial compared to imipramine. Hence, the present disclosure synthesizes a low cost norclomipramine loaded unilamellar liposomes as a norclomipramine delivery system for commercial purposes.
